Established in 1981, the Thirumuruga Kirupananda Variyar Thavathiru Sundara Swamigal (TKVTSS) Medical Educational and Charitable Trust is the source of Vinayaka Mission's Research Foundation. In 1982, Dr. A. Shanmugasundaram, the visionary Founder-Chairman, established the Vinayaka Mission's College of Pharmacy at Salem by building upon this foundation. Vinayaka Missions made history in the field of education with this establishment.

A major milestone was reached in 2001 when Vinayaka Missions was granted the esteemed designation of "Deemed to be University." Under Section 3 of the UGC Act, 1956, the Ministry of Human Resources Development, Government of India, granted this recognition with the support of the University Grants Commission (UGC). This award recognised the school's steadfast dedication to academic quality, its achievement of the highest standards, and its provision of first-rate facilities, all of which combined to establish its preeminence in the field of education. This significant event signalled Vinayaka Missions' transition into Vinayaka Mission's Research Foundation (VMRF), establishing the university as India's 48th.

The university's academic programs are approved by the appropriate regulatory bodies and universities, which attests to their caliber and applicability. The university has gained recognition on a national level, as evidenced by its placement in the University Category of the National Institutional Ranking Framework (NIRF) 2023, namely in the 151-200 range. Furthermore, VMRF(DU) has an A accreditation from the National Assessment and Accreditation Council (NAAC).
